Design & Optimization of Novel PROTACs, Glues, and Degrader Conjugates

From bifunctional degraders such as PROTACs, to monovalent molecular glues, degrader conjugates, and emerging proximity-based modalities, scientists are learning how to redirect the ubiquitin-proteasome, lysosomal, and autophagy machinery to eliminate drug targets once considered “undruggable.” They are exploring new chemistries, broader chemical and target space, novel ligases, and next-generation screening strategies and AI models to understand and predict whether the cellular interactions are selective, efficient, and therapeutically meaningful.

This two-part Degraders & Molecular Glues conference brings together leaders across medicinal chemistry, chemical biology, and drug discovery to examine what is working, where the biggest challenges remain, and how targeted degradation and induced proximity are evolving into viable therapeutic strategies.

Coverage will likely include:

  • Update on emerging PROTAC and molecular glue modalities and conjugates
  • Rational design and optimization of protein degraders and glues
  • Emerging covalent chemistries to induce proximity and degradation for pursuing new target space
  • Optimization of physicochemical properties and improving oral bioavailability and tissue specificity
  • Translational PK/PD modeling for efficient study design and efficacious dose projection
  • Structural and mechanistic characterization using innovative assays and platforms
  • Beyond CRBN/VHL: new ligases, cellular pathways, and mechanisms for targeted degradation
  • Overcoming drug resistance, safety, and toxicity concerns surrounding protein degraders
  • Pursuing extracellular as well as hard-to-drug intracellular targets for degradation
  • Use of AI/ML to design novel degraders/glues and identify new drug targets

The deadline for priority consideration is October 5, 2026.

All proposals are subject to review by session chairpersons and/or the Scientific Advisory Committee to ensure the overall quality of the conference program.
